چکیده

Stress conditions and proinflammatory cytokines activate the c-Jun NH2-terminal kinase (JNK), a member of the stress-activated group of mitogen-activated protein kinases (MAPKs). We recently demonstrated that inhibition of JNK signaling with the use of the islet-brain (IB) 1 and 2 proteins prevented interleukin (IL)-1 –induced pancreatic -cell death. Bioactive cell-permeable peptide inhibitors of JNK were engineered by linking the minimal 20–amino acid inhibitory domains of the IB proteins to the 10–amino acid HIV-TAT sequence that rapidly translocates inside cells. Kinase assays indicate that the inhibitors block activation of the transcription factor c-Jun by JNK. Addition of the peptides to the insulin-secreting TC-3 cell line results in a marked inhibition of IL-1 –induced c-jun and c-fos expression. The peptides protect TC-3 cells against apoptosis induced by IL-1 . All-D retro-inverso peptides penetrate cells as efficiently as the L-enantiomers, decrease c-Jun activation by JNK, and remain highly stable inside cells. These latter peptides confer full protection against IL-1 –induced apoptosis for up to 2 weeks of continual treatment with IL-1 . These data establish these bioactive cell-permeable peptides as potent pharmacological compounds that decrease intracellular JNK signaling and confer long-term protection to pancreatic -cells from IL-1 –induced apoptosis. Characterization of a novel JNK (c-Jun N-terminal kinase) inhibitory peptide.

An improved understanding of the roles of protein kinases in intracellular signalling and disease progression has driven significant advances in protein kinase inhibitor discovery. Peptide inhibitors that target the kinase protein substrate-binding site have continued to attract attention. In the present paper, we describe a novel JNK (c-Jun N-terminal kinase) inhibitory peptide PYC71N, which i...

The Role of Caspase 9 during Programmed Cell Death in Ciliary Ganglia of Chick Embryos

During programmed cell death (PCD) apoptosis is controlled by many factors such as proteases. With no specific protease (s) known during PCD in the developing nervous system so far, we sought to determine if any specific protease (s) is involved in this process and therefore used different protease inhibitors during PCD (from embryonic day 6 to 10) in chick embryos. Among the inhibitors commerc...

Celecoxib, indomethacin and ibuprofen prevent 6-hydroxydopamine-induced PC12 cell death through the inhibition of NFκB and SAPK/JNK pathways

Objective(s): The possible action of nonsteroidal anti-inflammatory drugs (NSAIDs) in the reduction of reactive oxygen species (ROS) and also as anti-apoptotic agents may suggest them as putative agents for the treatment of neurodegenerative diseases. This study was designed to explore some pathways alterations induced by NSAIDs following 6-hydroxydopamine (6-OHDA)-ind...

Identification of a Novel Tumor-Binding Peptide for Lung Cancer Through in-vitro Panning

Tumor-targeted therapies are playing growing roles in cancer research. The exploitation of these powerful therapeutic modalities largely depends on the discovery of tumor-targeting ligands. Phage display has proven a promising high throughput screening tool for the identification of novel specific peptides with high binding affinity to cancer cells. In the present study, we describe the use of ...

Cyclooxygenase inhibitors combined with deuterium-enriched water augment cytotoxicity in A549 lung cancer cell line via activation of apoptosis and MAPK pathways

Objective(s): Combination chemotherapy is a rational strategy to increase patient response and tolerability and to decrease adverse effects and drug resistance. Recently, the use of non-steroidal anti-inflammatory drugs (NSAIDs) has been reported to be associated with reduction in occurrence of a variety of cancers including lung cancer. On the other hand, growing evidences suggest that deuteri...
